So, this entire episode of Outlander basically revolved around an evil snake (Lord Voldemort, is that you?) biting Jamie's thigh, and it was all extremely dramatic and angsty so let's get right to it.
Jamie didn't cheat on her.
Just when it looked like Jamie might lose his leg or die, he survived and didn't require an amputation. The storyline is taken from the Diana Gabaldon novel The Fiery Cross which plays out in the same way. Ultimately, Jamie is fine and doesn't die, but it does give him a deeper sense of his mortality.
In a shocking turn of events, Outlander season 6, episode 6 saw Claire's apprentice Malva accuse Jamie of infidelity, claiming her unborn child is Jamie's, but the aftermath forgets one critical detail from Outlander season 3.
Why did Jamie have bite marks? Unfortunately, this revelation came for Jamie thanks to a prostitute, who left some bite marks on his thighs. But nothing happened, he swears! He was tempted, but he did not participate in any number-related activities with any women who were not Claire.
Brianna Invents a Leg-Saving Syringe.
If you're wondering whether Jamie's leg gets amputated, the answer is, thankfully, no. And it's all because Brianna ingeniously invented a syringe using a hollowed-out snake tooth.

Director Brendan Maher told Vulture that while both Menzies and Heughan had stunt doubles for the physically demanding sequence, neither used them much. “We had people for safety, just in case, but the nature of the work is that you want the actors to do as much as they possibly can,” Maher said.
Among these is one moment in season three when Jamie Fraser (played by Sam Heughan) slept with Mary McNab (Emma Campbell-Jones). The scene took place after Claire Fraser (Caitriona Balfe) returned to the future shortly before the Battle of Culloden after Jamie feared for the safety of his wife and their unborn child.

Jamie does admit he's slept with someone else, telling her about his one-night tryst with Mary MacNab (Emma Campbell-Jones) before he turned himself in to authorities following the Battle of Culloden nearly 20 years prior.
Some think Jamie's spirit (but not his body) traveled to through time to guide Claire to him; others suggest Jamie had a near-death experience after the Battle of Culloden, and that's why his ghost is 25 when he appears to Frank.
Sunday night's episode of Outlander, “Of Lost Things,” featured an intense and intimate sex scene between Jamie Fraser (Sam Heughan) and Lady Geneva Dunsany (Hannah James). She essentially blackmailed him into sleeping with her by threatening to expose his true Jacobite rebel identity to her British family.
Jamie denies having slept with her, but the damage to his and to Claire's reputation is done—and the bond between the Frasers and Malva is severed, until just a few weeks later, when Claire finds Malva murdered in the garden.
The day after their wedding, John gives Claire a large chest of medical equipment as a wedding present. Claire, remembering a similar anniversary gift Jamie had given to her a decade earlier, faints. A week after her marriage to Lord John, she and John drunkenly sleep together.
Yet, even with so many people suspicious of her, there are plenty of characters who know that Claire is a time traveler. As the number of time travelers on Outlander increases, the secret understandably has become harder to keep. But the more people who know, the more Claire and her family could be in danger.
Throughout Outlander, Claire was seen wearing two wedding rings – both when she married her second husband Jamie and when she returned to the future and reconciled with Frank Randall (Tobias Menzies). Claire steadfastly refused to take off the gold wedding band from Frank after travelling back through time.
It helps to remember the whole timeline, which is easier when you read the books. When Jamie met /married Claire, he was a 23 year old virgin, and they were together three years before she went back through the stones pregnant with Brianna in 1746.

However, after some convincing from his sister Jenny Murray (Laura Donnelly), Jamie married Laoghaire. Sadly, their relationship didn't work out and before long she moved out with the pair estranged.
After the dust has settled, and with assistance from Ned Gowan, Jamie's marriage to Laoghaire is declared invalid – as Claire was clearly still alive – and he agrees to pay her a yearly fee and provide a dowry for each of her two daughters.
The simple answer is no. Claire and Jamie don't have any more children after Brianna. Claire heads through the stones while pregnant with Brianna at the end of the second season/book. She spends 20 years in the 20th century, and she and Frank don't have any children together.
