Well Clone Wars season 7 shows us that the time from battle of Coruscant to order 66 was just as long as the Mandalorian invasion, and Anakin went to Mustafar right after order 66. The next day Padme went to Mustafar, got choked, and gave birth.

In Legends, Luke and Leia do eventually discover their mother's identity during the Swarm War Trilogy.
Vader agreed to help the handmaiden, silently recalling the time he spent with Padmé in the process. In this moment, he was once again Anakin Skywalker, a young man in love who simply wanted to change the world for the better.
This is confirmed later in the movie when Obi-Wan informs Padme that Anakin killed them. Both are torn apart with hurt and confusion, trying to make sense of something so terrible.

The real answer is: When the original trilogy was written and filmed, Anakin Skywalker was not the creator of C3PO. There was no consideration given to who Vader would recognize, because there was no reason for Vader to recognize the droid, at all.
