In Stranger Things, 001 (Henry Creel/Vecna) killed the Hawkins Lab children and staff because he felt superior, viewing them as weak imitations and resenting Dr. Brenner for enslaving people with powers; he wanted to eliminate them to prove his dominance and escape control, taking back the powers he felt were stolen and eventually becoming the Mind Flayer's (Flayer) minion.
The motive for One to kill everyone at Hawkins Lab was the belief that Dr. Brenner's research was enslaving the superior beings. While One clearly has issues, he raised the question we've long wondered – do we like Brenner?
