Over a century ago, Doma held the position of Upper Rank Six ( 上 じょう 弦 げん の 陸 ろく , Jōgen no Roku?), and during that tenure, he turned Gyutaro and Daki into demons and introduced them into the Twelve Kizuki.
When he became 20 he met Muzan Kibutsuji who transformed him into a Demon. As a Demon, Doma becomes stronger by feasting on many, many female humans who, according to Doma, better to eat because they have many nutrients inside them that are used to nurture babies.
The only reason he did was in hopes that one demon would become powerful enough to withstand sunlight. If Muzan were to consume such a demon, he would gain the power for himself and achieve his dream of becoming the perfect being.

Unlike many other demons, Muzan wasn't directly involved in turning these siblings into demons. Instead, it was Doma, the Upper Moon Two, who turned these two into flesh-eating monsters.
We first see Doma when he is still the Upper Moon Six while he is in the middle of eating a geisha. Later he becomes even stronger and rises to the position of Upper Moon Two. He dies because of the lethal amount of poison he absorbed through Shinobu Kocho's body followed by Kanao Tsuyuri beheading him.

In her final moments, she apologized to Inosuke for failing to protect him and tosses him into the river as an attempt to save his life. She was ultimately killed and eaten by Doma, who claimed to have devoured her to the bone.
The curse prevents Muzan's name from being spoken aloud, and any who do will meet a horrible demise.
At the age of 12 her success all came to an end when she stabbed the eye of a samurai while her brother was away working. Since her brother wasn't present to protect her, she was then tied up and burned alive and left to die in a ditch. Her brother then carried her burnt body around town searching for help.
Because of his desire to become stronger and Doma's strength allowing him to surpass himself to become Upper Two, Akaza harboured an intense hatred of Doma. Doma, as apathetic as he is, considered him his "best friend", seemingly ignoring the contempt Akaza possessed.
The only Demon he despises the most is the Upper Rank Two: Doma. It is quite a shocker that the second most potent Demon in the Demon Slayer is not Muzan's favorite.
It is revealed that Muzan's true fear of dying and immense desire to live and survive was due to him being born "dead," where his heart would constantly stop in his mother's womb and was assumed stillborn until he fought to breathe before his cremation.

Doma also displayed what arguably is his first true emotion, love, when he proclaimed his love for Shinobu after finding her cute.

As a human, Daki was known by the name 'Ume' instead. This was a rather peculiar choice for a name, especially since Ume was the disease that killed Daki's mother! While the death of Daki's mother was anything but tragic — given how abusive she was — his name does hint towards the manner of her demise.

Doma met Kotoha when she was roughly 18 years old, on a cold snowy night. She came to him while carrying her baby, and pleaded for him to save them from her abusive husband and mother-in-law. As a result, Doma killed her pursuers, and accepted Kotoha and her baby into his temple.
