Just because he has a healing factor doesn't mean he can't lose; it simply means his opponent has to get creative to find a way to keep him down. And quite a few opponents have managed to do just that. Since Deadpool knows he's a comic book character, he doesn't take most fights too personally.
While Deadpool is virtually unkillable due to his healing factor, he is not invincible. In fact, he's been beaten, maimed, and dismembered countless times in the comics and movies. Deadpool's immortality only extends to his ability to regenerate from almost any injury.

Given the latter ever fights Thanos with Infinity Gauntlet and all the Infinity Stones, The Mad Titan can easily erase Wade from existence, ending the fight in an instant.
Deadpool's healing factor means he can regenerate from almost any wound, but the Anti-Regeneration Ray incinerates its target in a way that prevents the activation of any healing factors (Wade can actually survive normal forms of incineration, as shown when he was killed by Thor.)
Cows, Clowns and Other Phobias
The fast-talking gunman seems almost fearless on the battlefield, but in reality, there's a lot that scares the Merc with the Mouth. There are two things though, that scare him above all: cows and clowns are a Deadpool weakness.
Deadpool has bovinophobia, which is the fear of cows. According to him, cows "scare the *#$% outta" him, as he believes their stare to be chilling, like they're "waiting."

He is Deadpool's archenemy for many issues and almost everything that happened to Deadpool was a part of an elaborate plan orchestrated by T-Ray.
Can Deadpool Survive A Nuke Deadpool can survive a nuclear bomb — how hard could it be, right? The merc has been known for being indestructible, but you would have never guessed that he could survive his very atoms being blown to bits.
Deadpool used Pym Particles to get a bunch of explosives in and then blew them all up, killing Captain America, Spider-Woman, Iron Man, and more.
Deadpool's immortality lies in his cancer and regeneration ability. According to the comics, cancer triggered his healing powers. As a result, the cancerous cells regenerate just like the normal, healthy ones do.
Unlike Wolverine, whose wounds sometimes cause excruciating pain as they heal depending on the severity, Deadpool has some degree of pain insensitivity.
Deadpool's Strength Feats

Typically speaking, if Wolverine had an arm chopped off, he would not re-grow a new arm. Deadpool's powers do, in fact, do that.
Deadpool has died many times, and being hurled into the sun would definitely end up being one of those times. Direct immersion in the Sun would kill nearly any protagonist in the MCU short of Ghost Rider and maybe the Silver Surfer.
Deadpool wouldn't be able to touch flash and flash wouldn't be able to kill deadpool. It's a never ending fight, if there was any fighting at all.
In the Invincible comics, Wade was indeed thrown into the burning Sun and is a proven way to kill Deadpool.
Yet, Deadpool couldn't defeat Hulk and there was even an instant where Hulk utterly dismembered Deadpool. The only way the Merc with a Mouth was able to get the upper hand was by not eliminating the Hulk, but instead terminating Bruce Banner.
Out of jealousy and spite, Thanos cursed Deadpool using a combination of science and necromancy to make him literally immortal. So, even if his entire being were vaporized down to the last molecule, he'd just respawn somewhere randomly, so he could never see death again.
He can't Starve to death. In one comic he was stuck under a fallen building (that's what I think it was), and he stayed alive in there for a long time.

He never truly wins. In a battle, Deadpool has no weapons to kill Doomsday with, and Doomsday would crush him. However, Deadpool would regenerate, and the battle would go on forever. If Deadpool did pull out a Anti-Doomsday weapon from his magic sac, Doomsday will return.
