Ocrevus (ocrelizumab) can increase the risk of certain cancers, particularly breast cancer, as observed in clinical trials where a small percentage of patients developed it compared to placebo, though the mechanism isn't fully understood. While other cancers like bladder, brain, and cervical cancer have been linked to MS itself and immunosuppressants, Ocrevus's specific contribution, especially to non-breast cancers, needs more research, though it's important for patients to follow standard cancer screenings like mammograms.
Another possible serious side effect of OCREVUS and OCREVUS ZUNOVO is an increased risk of cancers (malignancies), including breast cancer. In 3 main clinical trials, 6* out of 781 females (<1%) were diagnosed with breast cancer.
OCREVUS can cause infusion reactions that can be serious and require you to be hospitalized. OCREVUS increases your risk of getting certain infections, which can be life-threatening or cause death, including upper respiratory tract infections, lower respiratory tract infections, skin infections and herpes infections.
In clinical studies of patients who have been on Ocrevus for up to 10 years, 77% with relapsing MS were free from disability progression, and 92% continued to walk without needing assistance.
Overall, there is general agreement that immunosuppressant therapies, such as azathioprine, methotrexate, cyclophosphamide, and mitoxantrone have led to increased cancer risks in MS patients and this risk is related to patient's family history of cancer, duration of treatment and cumulative dose (16, 17).
People with MS had an increased risk for bladder cancer, brain cancer and cervical cancer. People with MS had a lower risk of prostate cancer, colorectal cancer and breast cancer. Cancer risk was higher in people with MS younger than 55 years and decreased in people with MS 65 years and older.
About 90% of cancers are caused by environmental and lifestyle factors, not genetics, including smoking, poor diet (red meat, fried foods), alcohol, sun exposure, pollutants, infections, obesity, and inactivity; only 5–10% are due to inherited genetic defects, with most cancers arising from lifestyle-induced genetic mutations. Tobacco alone accounts for about a third of cancer deaths, while diet, obesity, and inactivity contribute significantly, with controllable factors being key to prevention.
Patients treated with ocrelizumab were at significantly greater risk of serious infection compared with those who received platform injectable therapies (odds ratio = 1.98; 95% CI, 1.52-2.59; P < . 01). The rate of serious infections per 100 patient-years was 4.21 with ocrelizumab versus 2.64 with platform injectables.
Research Insights About MS and Lifespan
The study, which involved about 34,000 people, including 5,800 with MS, showed that people with the disease lived, on average, to be 75.9 years old. Meanwhile, those without MS lived to an average age of 83.4 years old.
Ocrelizumab (Ocrevus) was a lot better than beta interferon at reducing relapses. It was also much better at cutting the number of lesions seen on MRI scans. This means that in the two Opera trials, on average, people saw relapses drop by almost half over two years.
If you think you have become pregnant while receiving this medicine, tell your doctor right away. This medicine may increase your risk of developing serious infections (eg, bacterial, fungal, parasitic, viral). Avoid being near people who are sick or have infections while you are using this medicine.
Natalizumab scored highest for “availability of long-term safety data”; and cladribine tablets for “low risk of malignancy”, “good benefit vs. risk profile”, “good short- and long-term safety profile”, and “low risk of PML”. Ocrelizumab scored highest for being “relatively well tolerated”.
About half of people with MS at some time or other get a particular problem called optic neuritis. This is when your optic nerve becomes inflamed. Optic neuritis symptoms can include blurry vision, 'washed out' colour vision, and eye pain for a few days when you move your eyes.
Serious side effects reported with Ocrevus include:
Though less common, some patients with MS may develop uveitis, an inflammation of the uvea, the middle layer of the eye. This can cause redness, eye pain, light sensitivity, and blurred vision. Chronic or recurrent uveitis can lead to more severe complications, such as cataracts or glaucoma, if not treated promptly.
MS can weaken the muscles that control the lungs. Such respiratory issues are the major cause of sickness and death in people in the final stages of MS. Spasticity or an increase in stiffness and resistance as a muscle is moved can impair movement and cause pain and other problems.
Personal Independence Payment (PIP)
You can spend it on whatever you need, such as paying for support to remain independent during relapses, or to help with extra costs such as heating, transport or help around the house. Some people with MS assume they can't get PIP because they're 'not disabled enough'.
Multiple Sclerosis Final Stages: Symptoms
Balance and Coordination Issues: Standing, walking, and even sitting upright can become challenging. Severe Muscle Spasms and Stiffness: Cramping, stiffness, and painful spasms in muscles may become more frequent.
An increased risk of malignancy with ocrelizumab may exist. In controlled trials, malignancies, including breast cancer, occurred more frequently in OCREVUS-treated patients. Breast cancer occurred in 6 of 781 females treated with OCREVUS and none of 668 females treated with REBIF or placebo.
This ability will be further compromised if the person is also taking other drugs that suppress the immune system, such as chemotherapy agents, immune-modulating therapies, or other immunosuppressants. This immune-suppressing effect will persist for a few months after stopping Ocrevus treatment.
Weight gain and weight loss weren't reported as side effects in clinical studies of the drug. Keep in mind that Ocrevus may cause edema (swelling) in the arms or legs. This can lead to a small amount of weight gain. If you have edema with Ocrevus, talk with your doctor about ways to ease this side effect.
Cervical cancer is one of the most preventable cancers thanks to the HPV vaccine. HPV, or the human papillomavirus, causes 99% of all cervical cancers. The HPV vaccine, which is given in 2 doses, targets 9 different types of the virus.
Lung and bronchus, colorectal, pancreatic, and breast cancers are responsible for nearly 50% of all deaths.
Lycopene was found to inhibit human cancer cell proliferation, and to suppress insulin-like growth factor-I-stimulated growth.
